What is the most important information I should know about Avutometinib And Defactinib (Oral)?

Tell your doctor if you have or have ever had:

  • a history of vision or eye problems;
  • severe skin reaction; or
  • liver disease.

Avutometinib and defactinib can cause the breakdown of muscle tissue, which can lead to kidney failure. This happens more often in women, in older adults, or people who have kidney disease or poorly controlled hypothyroidism (underactive thyroid).

Avutometinib and defactinib can harm an unborn baby if the mother or the father is using this medicine.

  • If you are a woman, you may need a pregnancy test to make sure you are not pregnant. Use birth control while using this medicine and for at least 1 month after your last dose.
  • If you are a man, use birth control if your sex partner is able to get pregnant. Keep using birth control for at least 4 months after your last dose.
  • Tell your doctor right away if a pregnancy occurs.

Do not breastfeed while using this medicine, and for at least 2 weeks after your last dose.

Avutometinib and defactinib may affect fertility in men or women. Pregnancy could be harder to achieve while either parent is using this medicine.

Side Effects

What are the side effects of Avutometinib And Defactinib (Oral)?

Get emergency medical help if you have signs of an allergic reaction: hives, difficult breathing, swelling of your face, lips, tongue, or throat.

Seek medical treatment if you have a serious drug reaction that can affect many parts of your body. Symptoms may include skin rash, fever, swollen glands, muscle aches, severe weakness, unusual bruising, or yellowing of your skin or eyes.

Avutometinib and defactinib can cause the breakdown of muscle tissue, which can lead to kidney failure. Call your doctor right away if you have unexplained muscle pain, tenderness, or weakness especially if you also have fever, unusual tiredness, or dark urine.

Call your doctor at once if you have:

  • eye problems--vision changes, dry eyes, seeing flashes of light or "floaters" in your vision, redness, swelling and itching of your eyes or eyelids, eye pain;
  • skin problems--acne, blistering or peeling of the skin, skin infection, itching, skin rash, dry skin, blisters or ulcers in your mouth, redness or swelling of your face, hands, or soles of your feet; or
  • liver problems--loss of appetite, nausea, vomiting, stomach pain (upper right side), tiredness, itching, dark urine, clay-colored stools, jaundice (yellowing of the skin or eyes).

Your cancer treatments may change, be delayed or permanently discontinued if you have certain side effects.

Common side effects may include:

  • tiredness;
  • nausea, vomiting, diarrhea, constipation, stomach pain or upset;
  • cough, shortness of breath;
  • pain and burning when you urinate;
  • swelling in any part of your body;
  • abnormal blood or urine tests; or
  • bone pain or muscle pain.

What drugs and food should I avoid while taking Avutometinib And Defactinib (Oral)?

Avutometinib and defactinib could make you sunburn more easily. Avoid sunlight or tanning beds. Wear protective clothing and use sunscreen (SPF 30 or higher) when you are outdoors.

Ask your doctor before taking an antacid, and take only the type your doctor recommends.

How to take Avutometinib And Defactinib (Oral)?

Use Avutometinib And Defactinib (Oral) exactly as directed on the label, or as prescribed by your doctor. Do not use in larger or smaller amounts or for longer than recommended.

Do not change your dose or stop taking this medicine without your doctor's advice.

Take avutometinib and defactinib with a meal.

Swallow the avutometinib capsule whole and do not crush, chew, break, or open it. Take the medicine at the same time.

Swallow the defactinib tablet whole and do not crush, chew, or break it.

If you vomit shortly after taking avutometinib or defactinib, do not take another dose. Take your next dose as scheduled.

You may need frequent medical tests and your cancer treatments may be delayed or discontinued based on the results.

You will be given other medications to help prevent serious side effects. Keep using these medicines for as long as your doctor has prescribed.

Store avutometinib and defactinib in their original container in the refrigerator, do not freeze.

Take avutometinib as soon as you can, but skip the missed dose and take your next dose at the regular time if you are more than 24 hours late for the dose. Do not take two doses at one time.

Take defactinib as soon as you can, but skip the missed dose and take your next dose at the regular time if you are more than 6 hours late for the dose. Do not take two doses at one time.
